General description:
Fibrohol is a preparation based on obeticholic acid — a highly selective agonist of nuclear FXR receptors located in the liver and intestines. Obeticholic acid regulates bile acid metabolism by reducing their synthesis from cholesterol, lowering intracellular bile acid concentration in hepatocytes, and enhancing their transport out of the liver.
These mechanisms decrease cholestatic load, reduce the toxic effect of bile acids on the liver, and improve hepatic functional indicators.
Recommendations for use:
Digestive system and metabolism.
Preparations for the liver and biliary tract.
Bile acids and their derivatives.
Main effects:
FXR receptor agonist (liver and intestine)
regulation of bile acid metabolism
reduction of bile acid synthesis from cholesterol
decrease of intracellular bile acid concentration in hepatocytes
enhanced bile acid transport and excretion from the liver
reduction of the total circulating bile acid pool
reduced toxic load on the liver
decreased inflammation and cholestatic stress
improved liver function
Pharmacological properties:
Obeticholic acid acts as a selective FXR agonist, suppressing CYP7A1 activity and reducing bile acid synthesis from cholesterol while increasing the expression of transport proteins responsible for bile acid efflux from hepatocytes.
This results in reduced intracellular accumulation of toxic bile acids, decreased cholestatic stress, and improved hepatic function.
FXR activation also affects inflammation and fibrogenesis, contributing to improved liver condition.
Composition per capsules:
Obeticholic acid — 5 mg
Dosage and Administration:
1 capsule once daily.
Liver function must be evaluated prior to initiation.
Initial dose: 5 mg once daily for the first 6 months.
Further dosage — as prescribed by a specialist.
Dosing Regimen:
Oral administration.
When used together with bile acid–binding resins —
obeticholic acid should be taken at least 4–6 hours before or 4–6 hours after the resin.
Contraindications:
hypersensitivity to obeticholic acid
contraindicated in children under 12 years
contraindicated during pregnancy and breastfeeding
Side Effects:
dizziness, palpitations
very common: abdominal pain, discomfort
